I obviously know Jackman/Wolverine in X-men.

Leo DiCaprio was supposed to star in Boogie Nights, but Titanic kept him from the role. Drew Barrymore was in the mix for Rollergirl.
Johnny Depp was supposed to star in Mr. and Mrs. Smith, but he decided to take a break from acting.
Tom Cruise was up for Edward Scissorhands, but he didn't want to play someone with facial scars.
Lisa Kudrow was Roz in the pilot for Frasier, but the producers wanted someone who could stand up to Fraiser - apparently she played the part like Phoebe. Something similar happed with Ray Romano in NewsRadio and Joe Rogan got the part.
Michelle Pfeiffer was originally cast as Clarice Starling in Silence of the Lambs. That was back when Gene Hackman was going to be Hannibal Lecter.

Sorry, Dougary Scott was to play Wolverine but he had to do MI 2.

Listening to Thomas Wilson (biff) talk about shooting BTTF with Eric stoltz is funny. He talks about how because the guy was being a tool to him outside of shooting (because Eric’s method style carried over)he was about to lose it. They had shot enough to where people were lining up their next gig and he knew stuff wasn’t coming out well and he got summoned for a meeting. He thought when Robert zemeckis called him in to let him know there were going to be major changes he thought he was getting fired. Instead it was a meeting to say we love what you’re doing but we have to let Eric go and we are going to extend shooting and reshoot stuff. Needless to say huge relief and the movie goes on the BR classic American cinema.

Bill Murray was the first choice to play Ted Stryker in Airplane but he wanted too much money.

Vincent Price and Christopher Lee both turned down the role of Dr. Rumeck in Airplane before Leslie Nielson got the part. Price later said it was the worst mistake he ever made in his career.

Pete Rose was originally cast for the role of Roger Murdock in AIrplane then the studio schedule filming for August and September so the part had to be recast.

Carrie Fisher was going to be Miss Scarlett in Clue but she took the role of Miss Star Wars in rehab instead.

Gig Young was the original Waco Kid in Blazing Saddles but his alcoholism got him replaced by Gene WIlder a few days into production.

Jean Claude Van Damme was the original choice to wear the Predator suit. He's all of 5'9 though and everyone else on set averaged like 6'3 and 250 pounds. Also he hated the suit and couldn't move for shite in it. So they got 7'3 Kevin Peter Hall to do it instead.

Saw a Clancy Brown interview where he said Sean Connery was originally offered the role of Connor MacLeod in Highlander. Connery read the script and didn't want the title role, but was interested in the Ramirez role. They weren't about to turn down Connery for their movie, so we got the Egyptian Spaniard with the Scottish accent.
